Protein-protein interaction is an important form of its function. However, since most protein-protein interactions are transient, it becomes more difficult to study these PPIs. The crosslinking method provides great help for today's interaction research and proteomics research, that is, capturing or freezing these transient contacts to study which proteins are involved and how they interact. In this method, crosslinking reagents are used to covalently bond the proteins together as they interact, and then the protein-protein complex is captured and studied. Common functional groups on crosslinkers can freeze transient interactions in place, or fix weakly interacting molecules in a sufficiently stable complex for better separation and characterization.

In vivo crosslinking can capture the interactions between proteins in their natural environment, thereby reducing false positive interactions and avoiding the loss of interactions during cell lysis. In vitro crosslinking is more practical for researchers who want to modify proteins, and In vitro crosslinking can better target specific crosslinking events, because many reaction conditions such as the concentration of reactants, the purity of the target protein, ion strength, temperature and pH can be controlled during in vitro research.
